CoR Innovation Mechanism of Action 

CoR is a sulfoxide, also called a sulfoxide, is an organosulfur compound containing a sulfinyl (>SO) functional group attached to two carbon atoms. It is a polar functional group. Sulfoxides have Electron-donor properties. Example of another sulfoxide is dimethyl sulfoxide (DMSO), a common solvent. In prokaryotes (bacteria and archaea) there are several different electron donors and several different electron acceptors. That electrons can enter the chain at three levels: at the level of a dehydrogenase, at the level of the quinone pool, or at the level of a mobile cytochrome electron carrier. These levels correspond to successively more positive redox potentials, or to successively decreased potential differences relative to the terminal electron acceptor. In other words, they correspond to successively smaller Gibbs free energy changes for the overall redox reaction Donor → Acceptor.

Flavoproteins have either FMN (flavin mononucleotide) or FAD (flavin adenine dinucleotide) as a prosthetic group or as a cofactor. are involved in a wide array of biological processes, including removal of radicals contributing to oxidative stress, photosynthesis, and DNA repair. They are some of the most-studied families of enzymes. About 5-10% of flavoproteins have a covalently linked FAD. Based on the available structural data, FAD-binding sites can be divided into more than 200 different types.

90 flavoproteins are encoded in the human genome; about 84% require FAD and around 16% require FMN, whereas 5 proteins require both. Flavoproteins are mainly located in the mitochondria, of all flavoproteins, 90% perform redox reactions and the other 10% are transferases, lyases, isomerases, ligases.

FAD can exist in four redox states, which are the flavin-N(5)-oxide, quinone, semiquinone, and hydroquinone. FAD is converted between these states by accepting or donating electrons.

The semiquinone (FADH·) can be formed by either reduction of FAD or oxidation of FADH2 by accepting or donating one electron and one proton, respectively. Some proteins, however, generate and maintain a superoxidized form of the flavin cofactor, 

FAD has a more positive reduction potential than NAD+ and is a very strong oxidizing agent.FAD plays a major role as an enzyme cofactor along with flavin mononucleotide.  Current research indicates that distinct isoforms exist in the cytosol and mitochondria. It seems that FAD is synthesized in both locations and potentially transported where needed.

FAD-dependent proteins function in a large variety of metabolic pathways including electron transport, DNA repair, nucleotide biosynthesis, beta-oxidation of fatty acids, amino acid catabolism, as well as synthesis of other cofactors such as CoACoQ and heme groups. Due to the importance of flavoproteins, it is unsurprising that approximately 60% of human flavoproteins cause human disease when mutated. There are decreased levels of FAD synthesis. Both of these paths can result in a variety of symptoms, including developmental or gastrointestinal abnormalities, faulty fat break-down, anemia, neurological problems, cancer or heart disease, migraine, worsened vision and skin lesions.
